      AGMATINE SULFATE– Agmatine is a ubiquitous naturally occurring molecule (Tabor and Tabor 1984). It is biosynthesized by decarboxylation of the amino acid Arginine. Agmatine is found in low amounts in many food stuff derived from plants, fish, and animal products. Animal studies demonstrate that the salt of Agmatine, Agmatine Sulfate, is absorbed by the gastrointestinal tract and distributed in the body (Molderings, Heinen et al. 2003). Agmatine initially attracted attention as an endogenous ligand at imidazoline receptors and α2-adrenoceptors (Li, Regunathan et al. 1994). Independent of binding to those receptors, Agmatine induces a variety of physiological and pharmacological effects exhibiting a great therapeutic potential.

      Agmatine is normally released endogenously in response to stress and inflammation, by supplementing with an exogenous source one can maximize its effect on recovery (2). When using Agmatine immediately after workouts, the nutrients digested in a post workout meal or shake can be more effectively delivered to the muscle cells for enhanced recovery and muscle growth.

      BENEFITS OF AgmaPURE !

      -AgmaPURE boosts muscle pumps
      Agmatine sulfate appears to boost muscle pumps in a couple different ways. First, Agmatine sulfate may improve nutrient partitioning, leading to more muscle glycogen (carbohydrates stored in muscle tissue) stored and consequently more water retained in the muscle. This can lead to a full look to the muscle and increase muscle pumps. Second, Agmatine sulfate also appears to increase nitric oxide production by acting as a competitive inhibitor of the enzyme nitric oxide synthase (Demady DR & Jianmongkol S, et al.). The nutrient partitioning effects of agmatine may be due to its possible ability to increase the insulin response to carbohydrates, and it may also have something to do with the increased blood flow to the muscle experienced with increased nitric oxide production. Agmatine sulfate may play a role in the hypothalamic control of luteinizing hormone and growth hormone release, leading to increased levels of these hormones (Zarandi M & Serfozo P, et al.).

      -AgmaPURE Decreases blood pressure and heart rate
      Agmatine as endogenous ligand of imidazoline and α2-adrenoceptors participates in the regulation of blood pressure and heart rate. The stimulation of imidazoline and/or α2-adrenoceptors will suppresses the norepinephrine and epinephrine that mediate a number of physiological responses to endogenous catecholamines such as hypotension, sedation, inhibition of insulin release, anti-lipolysis and platelet aggregation (Gonzalez et al., 1996, Zhao & Ren,2005). Also imidazoline and/or α2-adrenoceptors are reported to related to reduction in calcium influx and potassium efflux, reducing the rate of depolarization and the amplitude of action potential in the heart, through the inhibition the increase in the intracellular [Ca2+] by blocking L-type voltage dependent Ca2+ channel (Li et al., 1999) (Li et al.2000a, 2002a,b)
      Finally, the decrease in systemic blood pressure in rats and rabbits after intravenous administration of Agmatine was sensitive to the nitric oxide synthase (NOS) inhibitor L-NAME (Haulica et al.,1999), suggesting an involvement of nitric oxide (NO).

      -AgmaPURE has antinociceptive effects
      Agmatine binds to NMDA receptors and acts as an antagonist at NMDA receptor channels (Gadotti et al., 2006), and NMDA antagonists are known to block opioid withdrawal symptoms (Trujillo & Akil, 1994). Agmatine reversed the up-regulation of the NR1 subunit of NMDA receptors in the nucleus accumbens inmorphine dependence without changing NR1 subunit expression per se (Wang et al., 2011).

      -AgmaPURE Represses the tumor cell growth
      There is evidence that an impairment of intracellular agmatine homeostasis is crucially involved inmalignant cell proliferation in solid tumors and leukemia (Molderings et al., 2004; Molderings, 2006; Wolf et al., 2007; Haenisch et al., 2008). In vivo, in human colon cancer tissue agmatine content and in parallel the expression of arginine decarboxylase (ADC) were distinctly reduced compared with that in the normal tissue adjacent to the malignant tumor (Molderings et al., 2004). Decrease of intracellular agmatine content by RNA interference targeting ADC in human intestinal tumor cells resulted in a significant increase of proliferation (Molderings, 2006).

      -Potential diuretic
      Agmatine can be formed and metabolized in rat kidney (Lortie et al., 1996). Administration of exogenous agmatine caused an increase in urinary flow rate by increasing osmotic clearance and a cAMP dependent inhibition of urea absorption in the terminal rat inner medullary collecting duct, effects attributed to binding to imidazoline recognition sites (Smyth & Penner, 1995; Penner & Smyth, 1996; Rouch & Kudo, 2002). Agmatine also produced renal vasodilatation and increases in nephron glomerular filtration rate (Lortie et al., 1996; Schwartz et al., 1997). This effect was not altered by denervation (Lortie et al., 1996) and may be mediated via a constitutive NO synthasedependent mechanism, since it was abolished by the NO synthase inhibitors NG-monomethyl-L-arginine and nitro-L-arginine methyl ester (Schwartz et al., 1997).

      HICA -  Alfa-Hydroxy-isocaproic acid (HICA) is an end product of leucine metabolism in human tissues such as muscle and connective tissue. According to the clinical and experimental studies, HICA can be considered as an anti-catabolic substance. The present study investigated the effects of HICA supplementation on body composition,delayed onset of muscle soreness (DOMS) and physical performance of athletes during a training period.

      Beta Alanine
      For many years now research has continued to support the usage of this amino acid derivative for performance enhancement. Working in a similar, yet more efficient fashion as Citrulline Malate, Beta Alanine increases muscle carnosine levels allowing for improvement in multiple areas of physical performance (3,6,7). 

      By buffering hydrogen (H+) ions, it allows for an increase in aerobic and anaerobic performance especially muscular endurance in weight training and short and long duration sprinting (3,5,6,7). A 2009 study showed even an increase in peak sprint power output by over 11% (3). Another recent study published in the peer reviewed journal Amino Acids showed that supplementing with Beta Alanine was able to increase muscle carnosine levels by as much as 64%, and low doses still caused an elevation over 40% (7). Beta alanine has also been shown to prevent the drop in blood pH seen in high intensity exercise, which may be a significant contributor to its ability to improve exercise performance (8).

      Aside from improving muscular endurance, strength athletes that train in low rep ranges can also benefit from Beta Alanine supplementation to decrease fatigue and increase total training volume (30). When training primarily with low reps, users will see the most benefit later in the workouts, by being able to add sets and experience less drop off due to fatigue accumulation.

      TAURINE

      Taurine has many fundamental biological roles such as conjugation of bile acidsantioxidationosmoregulationmembrane stabilization and modulation ofcalcium signaling. It is essential for cardiovascular function, and development and function of skeletal muscle, the retina and the central nervous system. Taurine produces anxiolytic effect and may act as a modulator or antianxiety agent in the central nervous system by activating the glycine receptor. In addition, taurine influences electrolyte functioning by stabilizing electrolyte levels in the bloodstream during exercise.

      Taurine in the Body & Food:

      Quantitatively, taurine is a very important amino acid. It is particularly high in heart and muscle tissue1, but is also among the highest amino acid in a range of tissues and organs. Compared with other species, humans do not synthesize a lot of taurine in their bodies, leaving the diet as the major determinant of taurine levels2. Because taurine occurs predominantly in eggs, seafood and meat; vegetarians and vegans in particularly are at risk of deficiency3, 4.

      Functions of Taurine

      Taurines key roles within the body include bile salt formation, antioxidant protection, osmoregulation, membrane stabilization and regulation of muscle contractile function2. When it comes to use in bodybuilding and sports nutrition supplements, taurine is often singled out for its antioxidant, osmoregulation and muscle contractile regulation properties. But just focusing on these aspects of taurine function doesnt do justice to the versatility of this unique amino acid.

      Benefits of Taurine

      It is not uncommon for individuals who start taking taurine to experience an improvement in their digestive function and cholesterol levels, which is due to taurines role in bile salt formation5. One other common report is an improvement in cardiac function, particularly in individuals who are prone to cardiac irregularities, such as irregular heartbeats4. Because taurine is involved in the regulation of electrolyte and mineral flux in muscle tissue, individuals prone to exercise-induced muscle cramps often report symptomatic improvement after supplementing with taurine5. Taurine also has strong links to cardiovascular health, with one study linking the longevity of the Japanese people with the high amount of taurine in their diet coming from their high seafood consumption6. While another study found that taurine and omega-3 fatty acids had superior effects on blood lipid profiles when compared with omega-3 fatty acids alone7. Lastly there is considerable research indicating taurine can be beneficial for minimising its complications associated with diabetes8.

      Taurine & Cell-Volumisation

      Osmoregulation, as the name suggests, describes the process of regulating the osmoticpressure of fluid to prevent it from becoming too dilute or concentrated. The human body requires that the concentration of fluids within different tissues and body compartments be strictly regulated to maintain proper working function. Taurine is regularly touted as being an osmoregulatory substance, which is the more scientific term for cell-volumising, however, unfortunately there is very little in the way of research to support these claims for taurine, particularly during exercise10. One of the few studies undertaken in humans found that taurine, when compared with sodium, did not play a key role in regulating the changes in plasma osmolality that occur with endurance exercise. In contrast, there has been a lot of research confirming taurines osmoregulatory role in the brain2, however, this has little relevance to exercise.

      Taurine for Aerobic Exercise

      Taurine has shown some promise as an effective tool for promoting aerobic exercise capacity and enhancing recovery. A study of young sedentary college students found that supplementation with 6 grams/day of taurine led to a decrease in exercise-induced oxidative stress and enhanced exercise performance compared to controls11.However it appears these effects were mediated simply by an increase in plasma taurine levels versus muscle levels. This was confirmed by a later study conducted by researchers from the UK. In this study, supplementation with taurine resulted in a significant increase in plasma levels, but didnt change muscle levels12. Taurine supplementation also had no effect on the metabolic responses to 120 min of exercise at 60% VO2peak12. The other study worth mentioning is the original Red Bullenergy drink study. This study showed that consumption of the original Red Bull energy drink; containing 2g of taurine, resulted in an improvement in markers of cardiac function, such as stroke volume in experienced triathletes, cyclists and runners13. While there has been a study published in rats, which showed that supplemental taurine resulted in an increase in muscle taurine levels, they used a dose of taurine equivalent to 40-44g/day9, which is well above any reasonable dose for a human.

      Taurine for Resistance Exercise

      No corresponding investigation has been conducted in humans on the effects of taurine supplementation on the stimulus or response to resistance exercise. However, there are a series of studies in rats with one showing taurine decreased the oxidative stress response to eccentric exercise14, and another showing taurine supplementation increased muscle force production and protected muscle function following high-frequency muscle stimulation15. Yet another showed that taurine administration in rats prevented the increase in oxidative damage associated with 90-minutes of downhill running, which is largely an eccentric type of activity renowned for increasing oxidative stress and damage to muscle tissue16.

      Taurine for Recovery

      Taurine is a very versatile and unique amino acid, which is highly concentrated in a range of tissues including muscle and heart. While proper clinical human studies are still lacking, it reasonable to say based on the current evidence that taurine can potentially improve recovery from resistance exercise while also enhancing exercise capacity. Taurine supplementation can benefit a wide range of parameters from insulin function to digestive function and has shown no toxicity in studies. This makes it an ideal supplement to add to your training regime as it is likely to improve your performance as well as enhance your health from a more holistic standpoint.
